바늘꽃과 달맞이꽃 추출물의 항산화 활성 및 멜라닌화에 관한 효과

Abstract
This study was aim to examine the antioxidant effect and melanogenesis of Oenothera lamarckiana SERINGE(OLS) extract in cultured human skin melanoma cell(SK-MEL-3). After cultured SK-MEL-3 cells were treated with various concentrations of glucose oxidase(GO) for 3 hours, cell viability and melanogenesis such as tyrosinase activity and melanin synthesis were assessed. And also, antioxidant effect was measured by superoxide dismutase(SOD)-like activity and lipid peroxidation using ferric thiocyanate(FTC). In this study, GO remarkably decreased cell viability in the treatment of 50mU/mL GO for 3 hours. And also, the pretreatment of OLS extract for 2 hours remarkably increased cell viability which was decreased by GO. In the antioxdant effect of OLS extract, the OLS extract increased cell viability inhibited by GO, the decreased lipid peroxidation and SOD-like activity. While, OLS extract decreased melanin synthesis and tyrosinase activity after SK-MEL-3 cells were pretreated with 70ug/mL or 100ug/mL of OLS extract for 2 hours in these cultures. From these results, it is suggested OLS extract showed not only the antioxidant effect by the incresed cell viability on GO, the decreased lipid peroxidation and SOD-like activity but also inhibited activity of melanogenesis via the decreased melanin synthesis and tyrosinase activity in these cultures.
